lactose regulated bla, lacI ORF mutation

This device will express b-lactamase regardless of environmental lactose concentration. The lacI biobrick contains a mutation in the open reading frame of the lacI gene that replaces the third and fourth codons with stop codons. Consequently, there will be no lac repressor produced to bind lacO, leading to the expression of b-lactamase regardless of environmental lactose or glucose concentration.

This is observed using nitrocefin (b-lactamase chromogenic substrate). The color change in all environments tested indicates the presence of this mutation.
