lactose regulated bla, lacP deletion

This part will express b-lactamase regardless of environmental lactose concentration. The bla gene is regulated by a lacI biobrick with a mutation in the -10 sequence of lacP. Six bases were removed from the lacP -10 region, resulting in a nonfunctional promoter. The mutation was verified by sequencing.

Additionally, the sequence was verified by observing a lack of color change in an environment with high lactose concentration and low glucose concentration. The bla gene of this part is under the control of lacP, since it is nonfunctional, bla will never be expressed regardless of environmental lactose or glucose concentration (pictured).
