Lactose regulated bla

This device alters b-lactamase expression using the regulatory sequences of the lac operon.

Since the bla gene is expressed according to the lac operon's regulatory components (from lacI biobrick Bba_K4062014), b-lactamase will be expressed in environments with high concentrations of lactose and low concentrations of glucose. When treated with b-lactamase substrate nitrocefin, the expression of bla can be visualized through a color change.

The figure below shows this color change. When exposed to lactose and nitrocefin, the color of the culture changes to red within 15 minutes. Alternatively, when exposed to glucose the culture does not turn red. The first figure shows the color change when exposed to 10ug/ml nitrocefin and the second figure shows the color change when exposed to 1ug/ml nitrocefin.
