Scaffold (for SH3 and Leucine Zipper)
The scaffold is a protein with an SH3 ligand on N terminus while a Leucine Zipper domain on the C terminus. The SH3 ligand could bind the engineered EnvZ (BBa_K4061020) with a high affinity binding to its SH3 binding domain present on its C terminus. The Leucine zipper on the C terminus of scaffold can bind the Leucine zipper binding domain present on engineered OmpR. This interaction can bring the two proteins in close proximity with high affinity binding, thereby enhancing the rate of phosphotransfer in the system.
This system could potentially enhance the activity even between any two non-cognate partners. A scaffold brings two proteins together in a relatively stable configuration and enhances the signalling between two proteins given the close proximity and localisation.
