Engineered OmpR (facilitates scaffolding)
OmpR is a regulatory protein which controls the expression of OmpC and OmpF promoter belonging to the Two Component System of E. coli. This part contains the native CDS of OmpR regulatory protein in E. coli engineered to have a Leucine zipper binding domain at its C terminus. This addition allows binding to a scaffold (BBa_K4061015) which should enhance the rate of phosphotransfer from its cognate response regulator, EnvZ. The leucine zipper on the C terminus of the protein is attached with an inert linker chain of amino acids Glycine and Serine. This addition will not impact the function of OmpR if used without any of the other engineered components such as EnvZ or the scaffold protein. In order for an enhanced response of OmpR regulation, this part along with engineered EnvZ (BBa_K4061020) and the scaffold protein need to be used. Teams working with OmpC promoter should consider using this scaffold interaction as this can enhance interaction between the response regulators and speed up the regulation time.
