LasR codon optimized for B. subtilis
LasR is an allosteric trancription factor which originates from P. aeruginosa and regulates the gene expression of quorum-sensing genes. It binds the acyl homoserine lactone N-(3-oxododecanoyl)-homoserine lactone (3OC12-HSL). 3OC12-HSL is an autoinducer by which gene expression in P. aeruginosa is regulated. By forming a complex with 3OC12-HSL, LasR induces the promoter PLux. [1]
In our designed genetic circuit, the gene encoding the transcription factor LasR is controlled by a constitutive promoter originating from B. subtilis. The allosteric transcription factor binds the autoinducer 3OC12-HSL and thus its presence can be detected through eGFP expression.
