UAS_FEC -> upstream activating sequence
Protein expression ranges can be broadened by adding upstream activating sequences that bind transcription factors upstream of the promoter of interest.
Usage and Biology
In 2019, Decoene et. al constructed short, synthetic UASs increase TF binding before synthetic core promoters modified from TEF promoter UAS. Although UAS_TEF in front of some synthetic promoters did not improve expression, synthetic UAS_FEC significantly increased promoter activity for cpRedden1 and cpTEF_6-I by 4.8 and 2.5, respectively. The Part was added in front of cpTEF_6-I in CCA_San_Diego 2021 project in order to increase protein expression levels in yeast. Although it may not have the same effect, it may be used in front of any promoter to theoretically increase protein yields.
