cI repressor
Phage lambda forms the repressor cI, which controls the decision to initiate the lytic or the lysogenic cycle. This repressor binds itself to the O1, O2 and O3 operators, forms multimers and bends DNA, preventing transcription initiation from the pR promoter. We added one of the operator sequences into the Pgrac promoter to create Pgrac OcI which should then be repressed in the presence of the cI repressor.
