PDC1 promoter from S. cerevisiae
PDC1(BBa_K3772007) is a promoter that cloned from a coding sequence of enzyme that involved in Glycolysis pathway, as it is shown in figure, there are 3 binding sites inside ALD4p that allow transcription factor GCR1 to bind with them.
The fermentation results and corresponding yield of valencene when PDC1p is transferred into the S. cerevisiae are shown below.
The optimal promoters in each pathway were screened to investigate the relationship between glucose concentration, ethanol concentration and valencene yield with the extension of fermentation time, the results are shown in the figure below.
The next figure shows the fermentation results and corresponding yield of valencene when PDC1p is transferred into the S. cerevisiae, from which we know that PDC1p was high, and it was mainly expressed in the prophase (0-24h).
To further investigate and compare the 4 promoters, specific production rate of product is evaluated, and it shows that PDC1p function as a promoter that is responsible for production of prophase.
