Bacterial expression vector
This is a linear plasmid backbone that contains an origin of replication, chloramphenicol resistance sequence, ribosome binding site, and a 6xhis tag. We use it as the backbone for the sequences we wish to be translated. Our team gets this part from performing PCR on the plasmid which is originally used to be translated into SARS CoV2 spike protein(addgene id: 145730). In our project, we use this backbone for all of our protein coding sequences. For example, we enclosed the coding sequence of RFP, pepP protease, ACE2, Venus FP, and small fragments of a polypeptide. Most of them resulted in positive outcomes with the corresponding protein after protein purification(in this case, Ni-NTA his tagged protein purification kit of Qiagen). The result is proved by both SDS-page and Western blot assay.
