DNA Walker_lineL(for detection of N gene of SARS-CoV-2)
BBa_K3480001, what we called DW_lineL, is an oligonucleotide which used to build a nano machine called DNA Walker. DW_lineL is used to identify and bind potential viral genomes in the environment, thus promote the working of DNA Walker. Its 15 bases at the 3' end are paired with a conserved site in the N gene of SARS-CoV-2, while the other 25 bases at 5' end are all T. The oligoT is designed to help DW_lineL be linked to a solid carrier surface. For instance, we choose streptavidin magnetic beads as solid carrier to build our DNA Walker, than we need to modify a biotin at the 5' end of DW_lineL. The exist of oligoT can promote the flexibility of DW_lineL and reduce steric hindrance. DW_lineL can only work normally in DNA Walker. DW_lineL and DW_lineW will identify the 3' half and 5' half of the target sequence respectively.
