I1-60GFPHis - ATPase Inhibitor with GFP and His-Tag
The part consists of an inhibitor sequence (I1-60) that can bind to ATPase. It inhibits the hydrolytic function of the ATPase but not the ATP synthesis. The inhibitor sequence originates from a bovine inhibitor protein IF1. Only the first 60 amino acid residues of this protein are needed for this purpose. The inhibitor sequence is fused with a 17x helix to a eGFP and a His-Tag. The whole part can be used to purify a ATPase with a affinity purification via the His-Tag.
The construct is controlled by a Lac-Operon including LacI, LacO and a T7 promoter and terminator.
