CaFF-I

CaFF-I is the coding sequence of a fusion protein composed of two parts: intimin transmembrane domain and anti-caffeine VHH. Intimin is a protein found in enterohemorragic E. coli O157:H7. It is expressed and inserted in the outer membrane of the bacteria, exposing the C-terminal region to the extracellular medium. This exposure allows single-domain antibodies to bind to antigens in the media. In this case, a caffeine-binding nanobody is expressed, which in theory allows the bacteria to bind tightly to caffeine molecules.
