Part:BBa_K3425040
β-subunit of Qβ replicase
BBa_K3425040 is the β-subunit of Qβ replicase retrieved from the publication [1]. The sequence is adapted to Type IIS iGEM standard assembly (RFC1000) and therefore lacks the start codon. When assembled using Type IIS standard, the ATG start codon is included in the scar between RBS and coding sequence part. You can find more information about how to use this standard in the Type IIS guide of Uppsala 2020 team. Note, that the protein sequence is distinct from the β-subunit BioBrick BBa_K1906005.
About Qβ replicaseQβ replicase is an RNA-dependent RNA polymerase of Qβ phage. Functional enzyme consists of four parts, where only a single component - the β-subunit - is of viral origin. The remaining three components are ribosomal protein S1 and the translation elongation factors EF-Tu and EF-Ts. For its function, Qβ replicase requires the presence of specific recognition sequences at the both ends of its RNA template, such as QB5' (BBa_K3425038) and QB3' (BBa_K3425039) or MDV upstream and downstream.
