T7 strong promoter with ArsR binding site

The construct is comprising a T7 class III strong promoter (BBa_z0251) followed by an ArsR binding site. When the ArsR homodimer binds to the ArsR binding site, it represses transcription. The ArsR transcription factor as well as the ArsR binding site was identified in Escherichia coli str. K-12 substrain MG1655. When the ArsR dimer binds Arsenic, the ArsR homodimer unbinds the ArsR binding site, allowing the T7 RNA polymerase to bind and proceed with transcription.

Although it is very simmilar to the BBa_K190015 (more info about the function of the ArsR promoter can be found there), this construct has the ArsR binding site after the promoter region rather than preceding it (BBa_K190015). The reasoning behind this is the fact that although the ArsR dimer bound to the ArsR region represses transcription, it only represses it to the basal rate. To further strengthen the repression, this construct is supposed to obstruct the movement of the T7 RNA polymerae by being in its way.
