Pr-GFPmut3b-Term

Part BBa_K3252041 is the GFPmut3b coding sequence (BBa_E0040) that we used as reporter gene. This parts was constructed with BBa_K3252025 as RBS (BBa_B0034), BBa_K3252023 as (BBa_J23115 constitutive promoter), and BBa_K3252034 transcription terminator (BBa_B0015). That parts were assembled by order custom gene synthetic.

By using our experimental protocol, we characterize this expression level which is growth in different level of salinity. We cultivate Escherichia coli TOP10 that carrying BBa_K3252041 in LB media with different level of salinity.

1. LB media with 10 ppt

2. LB media with 25 ppt

3. LB media with 35 ppt

4. LB media with 45 ppt

5. LB media with sea water

Different level of salinity will influence the cell growth and the level of expression of some gene. In this case expresssion level of GFPmut3b was influence by the cell growth or OD600 that we observed. See our result page for detailed information.
