Part BBa_K3252038 is basic composite part that express LuxN protein. LuxN protein is one of the phosphorelated protein that used in Qrr RNA Lux cascade system in Vibrio parahaemolyticus. LuxN will autophosporelated when present the consentration of AI is low. BBa_K3252038 was controled under p66 constitutive promoter (BBa_K3252021) and very weak RBS BBa_K3252027. BBa_K3252018 was assemble with thats part and BBa_K3252034 transcription terminator by order custom gene synthetic.
LuxN protein will autophosporelated when the cell density is low (present of AI is low) and will be dephosporelated when the cell density is high (present of AI is high). Phosporelated LuxN will intereact with LuxU and activated LuxU (phosporelated). Activated LuxU will activated LuxO by phosporilating LuxO. Phosporelated LuxO protein will regulate the pQrr4 promoter that will express gRNA which consist of gRNAJ23115 and dCas9 handle (BBa_K3252013) as CRISPRi inverter element.
Characterization of this part was perform by full system integration BBa_K3252044 transcription unit.
