CRISPR-X: deactivated Cas9 coupled to activation induced deaminase (AID)
A gene encoding a mutant MS2 coat protein (BBa_K3218006) with a nuclear localization signal fused to an Activation-induced Deaminase (AID). The gene also encodes a dCas9 protein used to target specific loci within DNA. MS2 protein is used to attract the MS2-AID complex to the hairpin loops on the gRNA. In this manner the AID is brought to the target locus within the DNA. An improved version of MS2 protein is encoded which has a higher affinity for the MS2 hairpin loop.
