Blue light regulated inducer circuit
This circuit utilises the blue light sensor YF1 together with fixJ for detect the light, regulating the PhIF regulated by the promoter PfixK2 fixJ-regulated. The PhIF regulates the sigma fragment T3 which is part of the T7 RNAP core. We reconstructed this part for being able to induces only the blue light, independent from the red and green regulations, since the entire regulation is originally spread across different plasmids. This was constructed based in the parts from Jesus & Voigt et al 2017, that went characterised and submitted in the registry by the UCAS-China 2018 iGEM team.
Usage and Biology
Utilizing the plasmids from Jesus & Voigt et al 2017 we reconstructed only the blue light detector, therefore being able to be regulated in a single construct. This circuit can be used with the part BBa_K2598026 for induction controlled only by blue led light.
