Lpp-OmpA
Lpp-OmpA is an outer membrane protein that helps express the linked protein on the outer cell membrane. Lpp-OmpA is composed of two parts, lipoprotein and the OmpA domain. Lipoprotein can target to the outer cell membrane, which leads the whole protein binds to the membrane; and the OmpA domain is responsible for the expression of the linked protein.
Usage and Biology
To express lipase on the outer cell membrane, we modified and improved the submitted Lpp-OmpA by iGEM15_NCTU-Formosa (BBa_K1694002 https://parts.igem.org/Part:BBa_K1694002). In our design, we replace the original NcoI restriction site with restriction sites AvrII and NheI which brings some benefits to the future application of Lpp-OmpA. First, we found that NcoI is a restriction site on the plasmid, pSB1C3, a commonly used plasmid in iGEM. Therefore, with this modification, Lpp-OmpA is able to apply to common used plasmid in iGEM. Furthermore, with the two designed restriction sites, we can link various protein with Lpp-OmpA simply by adding AvrII and NheI restriction sites in front and behind the gene sequence.
