Modified 0.19 Wheat Albumin (Ile to Asn) expression using TDH3, RBS, secretion tag, and ADH3
This is a construct we used to express our modified 0.19 Wheat albumin, which acts as an alpha-amylase inhibitor. It can be transformed into yeast, and since there is a secretion tag, the 0.19 will be secreted into the solution or gel of the culture. Additionally, users should be aware that due to an amino acid substitution (the 44th amino acid, Isoleucine was changed to Asparagine), the attractions between amylase and the 0.19 were strengthened compared to the original 0.19 protein (hydrophobic to hydrophilic, thus lessening repulsion.)
