This part is a catechol 2,3-dioxygenase (CDO) expression cassette under the control of the Flavescence Doree 2.4 toehold switch for sequence-based detection of Candidatus phytoplasma vitis. It was specified with an RFC[10] scar between parts. The RFC[10] scar TACTAG between the switch and the downstream gene introduces an inframe stop codon and leads to premature termination of translation and thus no reporter production (for details see our characterisation of BBa_K2916068 a part from the same collection and of its improvement BBa_K3453170).
Moreover, the toehold switch sensor of this part has the trigger binding site is in the same orientation as in the transcriptional orientation of its target gene. Thus it is unlikely that this toehold switch will be able to detect the presence of an RNA naturally expressed in a cell.
