LsrA promoter-GFP
Long Description
This part is designed to detect whether the LsrA promoter activation is controlled by the quorum sensing signal via monitoring the GFP reporter gene expression.
Characterization
In this part of work, we constructed and transformed two plasmids into K12 E.coli strain for detecting lactic acid concentration. One plasmid includes the lldPRD operon promoter-LuxS-Lldr system that express the LuxS protein under the lactic acid induction, LuxS protein is critical for AI-2 quorum sensing protein production within the cell. As described previously, AI-2 protein enables to activate the LsrA promoter activation and induce its downstream GFP expression. Thus, we could detect the green fluorescence intensity under lactic acid induction. From the fluorescence intensity data, lactate induction showed positively regulation for GFP expression and the 1mM of lactate with the greatest signal intensity (Figure 1).
