pTet+RBS+GFP+RBS+TetR+TT
This part is an auto-negative transcriptional feedback control loop. Auto-negative feedback loops are extensively found network motif in biological networks due to their faster response times. It is pTet-TetR regulated auto-negative transcriptional feedback loop.
The pTet (repressible promoter) is repressed by TetR. TetR is expressed when transcription is able to take place normally. This creates a negative feedback loop. The higher the expression of the sequence by pTet promoter, the more will be the production of the pTet and consequently stronger repression of pTet by TetR. We have used GFP as reporter for the measurement of expression levels of the construct. The GFP expressed under the same promoter i.e. pTet allows us to visualize the effect of auto-negative feedback on the construct via fluorescence analysis.
