Second node_BxbI-Xis_Recombinase based IFFL
This part is another second node for our Recombinase based incoherent feedforward loop. It contains the BxbI recombinase directionality factor within the attB and attP sites of BxbI Integrase, followed by a constitutive P7 promoter in the reversed direction. Hence, it competes to reverse the recombination caused by the BxbI Integrase.
This works in tandem with our first node, where BxbI Integrase is expressed via induction, which in turn activates our second node and third node simultaneously, generating reporter protein as well as BxbI RDF(Recombination Directionality Factor), which upon interaction with BxbI Integrase performs attL x attR recombination on the recombinant sites of BxbI in the third node, effectively switching off the circuit, and providing a pulse response, thus completing our incoherent feedforward loop.
