PpetE

pPetE copper inducible promoter for use in the Cyanobacterium Synechocystis.

Usage and Biology

PpetE is the promotor for the Synechocystis petE gene encoding a copper dependent plastocyanin of the electron transport chain. When lacking copper the PpetE is silent and the cell utilizes other [Iron dependent] electron carriers.

PpetE functions as an inducible promoter in Synecocystis by keeping the growth medium copper free (ddH2O) and acid washing of glassware. The cells are getting by with trace amounts of copper and their backup electron carriers.

Induction is accomplished by adding copper back to the medium with a level indicated below:

Silent: ~0 M Cu2+

Induced: 3µM CuSO4 (about 3x levels of standard [BG-11] growth media)

Strength: Experience tells Medium-High Synechocystis promotor.
