Scaffolding protein 141 from P22 fused with Cas9 (SPCas9)
SPCas9 is a cargo for the P22 capsid protein from P22 bacteriophage. The truncated scaffolding protein is originally found in the P22 bacteriophage genome and it helps with forming P22 capsids. When fused with the Cas9 it can cause formation of the P22 around the Cas9, effectively encapsulating it [1]. This part has a hexahistidine tag on the N-terminus of the construct.
Figure 1- A 12% SDS-PAGE gel of the scaffold protein-Cas9 fusion using nickel affinity chromatography. The elutions show that the proteins were purified, where the protein is at the 245kDa.
