B0033m is a variant of B0033 designed for use in Moclo type assemblies (has added spacers on the side). B0033 is a weak RBS, from the Weiss series. B0033m was designed in Iverson et al. 2015.1

NYU Abu Dhabi 2024 Characterization

NYU-Abu-Dhabi 2024 BBa_B0033_m0 was assembled using Golden Gate Assembly, with eYFP as a reporter to measure its ribosome binding site (RBS) activity via fluorescence at two temperatures: 28°C and 37°C. In the RBS activity graph, BBa_B0033_m0 is represented by the green lines. At 37°C (solid green line), BBa_B0033_m0 started with low activity, which then gradually increased before stabilizing and slightly declining towards the end of the time course. At 28°C (dashed green line), BBa_B0033_m0 followed a similar trend, starting with low activity, then steadily rising and reaching a peak before decreasing slightly. The activity at 28°C was consistently higher than at 37°C, indicating that BBa_B0033_m0 performs better at lower temperatures than under heat stress. Figure 1.
