Applications of BBa_K2680522

iGEM 2019 Characterization

As we were interested in utilizing quorum sensing as a vital component of our project, we characterized BBa_K2680522 with our circuit BBa_K3059769. In order to test out how sensitive our receiver circuit is to LuxI HSL production, we decided to measure Fluorescence over time in a plate reader experiment after induction with varying concentrations of HSL, ranging from 1nM to 10uM.

In the graph below, each data point represents the average fluorescence / OD600 value of 5 individual datapoints. The error bars are +/- 2 standard errors. The y-axis uses a Log10 scale as fluorescence is being measured.


As can be seen in the graph, BBa_K2680522 has a very steep induction curve occurring between 10nM and 100nM. This is useful for us, and other iGEM teams to know, as this switch like behavior is useful for distance-dependent patterning.
