WM_Pad2

Orthoganol flanker sequence on the suffix side of all 3G Assembly parts from William_and_Mary iGEM 2018. Along with BBa_K2680500, this allows easy PCR of inserts for confirmation of colonies or for the generation of additional DNA for 3G assembly (without miniprep). Further, these WM_Pad sequences allow for high-quality PCR of iGEM backbones for use in Gibson Assembly. Both WM_Pad sequences are free of BsmBI and BsaI sites and so are compatible with most Golden Gate Assembly platforms. Additionally, the WM_Pad sequences are orthogonal to the E. coli genome and to the UNS sequences used in 3G assembly.
