siRNA Target Vector 2 with BFP and GGGGS Linker
This Vektor is used in our siRNA Testing System as a Target Vektor. It Produces a Target mRNA which also contains a Linker sequence of an GGGGS Linker and a LacI Inhibitor sequence. When the mRNA is not silenced, a Fusion Protein of the LacI inhibitor and the Geneproduct of the target mRNA is produced and silences blocks the pLac Promotor so that no Markerprotein is expressed. When the Target mRNA is silenced, no Repressor is Expressed and the Reporter Protein gets expressed. This Version of the Targetvektor 2 uses mTagBFP as a Marker Protein. The Target mRNA is transkribed using the pTet promotor. To make this promotor inducible in strains which do not produce TetR a TetR Expression casette is added in the reverse strand. mTagBFP has an excitation peak at 399 nm and an emission peak at 456 nm.
