Nanoluciferase with C/D box sequence and 4*VEEV RNA stabilizing sequence at the 3' UTR
C/D box-VEEV can be added to 3'-UTR region of actively loading RNA to bind to RNA binding domain of packaging device 1 (L7AE). VEEV would stable mRNA during transporting process and transcribing in target cells. By combining these two sequences on 3' UTR region of a target gene, this gene can be packaged into exosome and delivered to target cell and exist for more than 36 hours. The reporter gene in this part is Nano luciferase, which is a very small luciferase that is only 19kDa. by adding luciferase substrate, Nano luciferase will give off light that can be detected by enzyme-labeled instruments. Therefore this part is designed for researchers to exam whether the packaging device could work.
