glycosylation sequon (GNSTM)
GNSTM is a 5-residue sequon which acts as an attachment site for N-linked glycosylation. It is demonstrated that linkage of this sequon with an exosome-targeting protein (e.g. RVG-Lamp2b (BBa_K1180002)) can introduce an efficient glycosylation site that prevents the targeting peptide from degradation, without effecting the peptide-receptor interaction at targeted cells. In projects that use exosomes as therapeutic drug carriers, GNSTM can be fused along with a targeting peptide to an exosomal membrane protein for cell-type-specific uptake of exosomes. It can enhance the stability of exosome-targeting peptide, and lead to an increase in the drug-delivery efficiency of exosomes.
