pGapA
Promoter GapA is naturally occuring constitutive promoter in Escherichia coli .
Usage and Biology
During our project, we isolated the GapA promoter from the E. coli genome. In contrast to many synthetic promoters, this novel promoter provides an indication of high levels of natural expression. Therefore, this promoter can be used as a future positive control of reporter production - such as GFP - in a natural situation. Moreover, GapA being a natural occurring promoter means that there are still possibilities for optimization through radiation. This way variations of this promoter can provide access to an even wider spectrum of colors.
In our project, we combined this naturally occurring constitutive promoter both with the blue chromoprotein AmilCP and the pink chromoprotein spisPink, creating pGapA-AmilCP (BBa_K2610041) pGapA-spisPink (BBa_K2610042). These constructs allowed us to compare natural high expression to the responses of our novel stress promoter+chromoprotein constructs.
