KSI_DP-LL37-ELST-P2-ELST
Sequence codes a polypeptide consisting of ketosteroid isomerase (KSI) with additional Asp-Pro dipeptide followed by LL37 and two elastin like segments with alpha helical segment P2 inbetween. KSI facilitates isolation of protein since it causes packing of the polypeptide in inclusion bodies. KSI can later be removed by exposing the polypeptide to acidic environment which results in cleavage of D-P peptide bond. LL37 peptide has antimicrobial activity against bacteria, fungi, and viral pathogens. Elastin-like segments are comprised of 10 elastin-like repeats VPGVG each (hydrophilic domains which contain residues important for cross-linking into stable polymer elastin). This polypeptide could assemble in random network based on cross-linking of elastin segments. When similar polypeptide KSI-DP-LL37-ELST-P1-ELST is added to KSI-DP-LL37-ELST-P2-ELST, coiled coiles between P1 and P2 alpha helical segments could also form.
