Part:BBa_K245071
KSI-DP-P6-P4-P2
Sequence codes a polypeptide consisting of ketosteroid isomerase (KSI) with additional Asp-Pro dipeptide followed by three alpha helical segments. KSI facilitates isolation of protein since it causes packing of the polypeptide in inclusion bodies. KSI can later be removed by exposing the polypeptide to acidic environment which results in brakeage of D-P peptide bond. When KSI is removed this polypeptide has basically identical properties as P6-P4-P2 (BBa_K245070). P6, P4 and P2 are all heterodimeric parallel coiled coil forming segments. P6 forms coiled coils with P5 (basic part: BBa_K245122), P4 with P3 (basic part: BBa_K245120) and P2 with P1 (basic part: BBa_K245118). Along with P1-P3-P5 polypeptide this construct could form planar network.
