KSI-DP-P1-H1-P2
Sequence codes a polypeptide consisting of ketosteroid isomerase (KSI) with additional Asp-Pro amino acid sequence followed by three alpha helical segments: P1, H1 and P2. KSI facilitates isolation of protein since it causes packing of the polypeptide in inclusion bodies. KSI can later be removed by exposing the polypeptide to acidic environment which results in brakeage of D-P peptide bond. When KSI is removed this polypeptide has basically identical properties as P1-H1-P2 (BBa_K245062). P1 and P2 from seperate polypeptide chanes form heterodimeric parallel coiled coil and H1 segments from two polypeptide chains form homodimeric parallel coiled coil. These polypeptides are supposed to assemble in different geometric forms (cube, tetraeder, ...) and hexagonal or triangular networks, depending on the concentration of the polypeptide in the solvent. The length and flexibility of the linkers (Ser-Gly) does not allow P1 and P2 segments from the same polypeptide chain to form coiled coil.
