pPsiA from Agrobacterium tumefaciens
This part is the promoter region (0.4 kb upstream) of the PsiA gene of Agrobacterium tumefaciens str. C58 (Atu4744)
pPsiA is predicted to be repressed by the PsiR transcription factor (BBa_K2448006) which is inhibited in the presence of D-Psicose [1].
Usage and Biology
Our team used this pPsiA promoter together with the Agrobacterium tumefaciens PsiR transcription factor (BBa_K2448006) to build a Psicose biosensor (BBa_K2448025). The results show that pPsiA is functional in E. coli and that it behaved as predicted under and without induction.
Using the BPROM webserver [2], the predicted features of this promoter are: Threshold for promoters - 0.20 Number of predicted promoters - 1 Promoter Pos: 117 LDF- 3.46 -10 box at pos. 102 tgatatggt Score 42 -35 box at pos. 83 ttgacg Score 60 Oligonucleotides from known TF binding sites: rpoD16: TAGAGCAC at position 67 Score - 10
