pLsrA+14 + B0032 + GFP
This is the lsrA promoter followed by and RBS and GFP. This lsrA promoter is derived from the E. coli sequence described by Xue et al. [1] which was first used by the Tokyo 2011 team. However, the Tokyo team missed out the last 14 bases of the promoter which we believed to be important in allowing the LsrR repressor protein to bind to the promoter. These 14 bases have been added on to this part.
The lsrA promoter should be inhibited by the LsrR protein. The LsrR protein can be bound and inhibited by autoinducer-2 allowing the lsrA promoter to start transcription of GFP.
Usage and Biology
We gained no definitive results to show that this part works as expected. Due to time constraints it was not possible to investigate further.
