Strep-DigA-Middle Linker-Fok_i

This part unites some of our basic parts. The combination of a StrepTag, an Anticalin-Tag and the linked protein domain Fok_i makes it possible to purify and detect the function of the protein. The DigoxigeninA tag allows the coupling to an digoxigenin linked oligonucleotide.

Usage and Biology

This composite part is one part of our universal endonuclease and needs another composite part like BBa_K243024 to build a functional heterodimer. The DigA tag leads the part to DNA which is hybridized with an Digoxigenin labeled oligonucleotide. The middle linker creates a distance of 24bp between the DigA and the linked Fok_i protein domain. The StrepTag serves as purification tag for Streptavidin column purification.
