6x Poly-histidine Tag codon optimised for use in E. coli
We have codon-optimised the DNA-sequence encoding 6 adjacent histidine residues to be expressed in E. coli efficiently. A poly-His tag is utilized to bind it to immobilized ions on a chromatography affinity column (often nickel, cobalt or copper), so as to isolate from a mixed solution the tagged protein of interest. The tagged protein will bind to the immobilized ions at a much stronger affinity than other proteins, so that all except the protein of interest can be eluted from the column with a low salt buffer. The protein of interest is then eluted with a high salt buffer.
The His-tag is useful as it is small (so can be easily added to either the N or C terminus) and a widely used method of purification, so its use can be validated by many previous experiments.
