Repressible aiiA
The aiiA gene has 750 nucleotides and encodes for a protein of 250aa whose predicted molecular mass is 28,036Da. This aiiA gene encodes for the autoinducer inactivation enzyme A (aiiA) LVA tagged. aiiA enzyme catalyzes the degradation of N-acyl-homoserine lactones (AHLs) better well known ad quorum sensing autoinducers.
The aiiA gene is expressed under the pTet promoter whose inhibition takes place when tetR is expressed. tetR expression is controlled by pBAD promoter and therefore aiiA expression gets inhibited under the induction of arabinose. When arabinose is added to the medium, PBAD triggers the expression of tetR which leads to the inhibition of pteT and hence represses the expression of aiiA which is under the control of pTet. Therefore, if unactivated, the aiiA is constitutivelly expressed, a non fluorescent state is expected. When the system is induced (arabinose is added), aiiA is not expected to be expressed and therefore AHL is not degraded by aiiA. The results can be seen here.
