AIP-induced expression system optimized for L.lactis(RBS+AgrC+RBS+AgrA+P2)
The agr operon is a gene regulatory system dicovered in Staphylococci, which acts a significant role in quruom sensing. Specifically for our sequence, it is correlated with group one AIPs (autoinducing peptides) and optimized for Lactococcus lactis. Additionally, the gene is extracted from Staphylococcus aureus, and the unmodified version of sequence derives from BBa_I746100. Moreover, the RBSes, which are respectively anterior to agrA and agrC, were cited from B0034.
This part is group one AIP sensing and expression-inducing generator. AgrC protein is membrane-inserted signaling molecule (group 1 AIP) receptor. After forming a complex with AIP, the complex can phosphorylate protein AgrA located in cytoplasm. Subsequently phosphorylated AgrA can induce the P2-promoter-initiated expression.
(Note: This part is only specific to AIP-I. Wrong use of types of autoinducing peptide may elicit expression-inhibitory effect, for instance, AIP-II and AIP-III. Overexpression of AgrC and AgrA will cause toxic effects to cells by competitively consuming ATP.)
