nitrite reductase cytochrome c-type protein NrfB H89Y
Our part encodes NrfB, a subunit of the Nrf enzyme which reduces nitrite to ammonia. The Nrf operon is composed of 7 genes (Nrf A, B, C, D, E, F, G) and encodes the E.coli cytochrome c nitrite reductase, which catalyses the 6 electron reduction of nitrite (NO2-) to ammonia (NH3). NrfA encodes the catalytic NrfA homodimer, which will form a heterotetrameric complex with the homodimeric NrfB gene product which facilitates electron transfer from the membrane-bound NrfCD complex. This part contains a point mutation that encodes an amino acid substitution (histidine to tyrosine) at position 89. This part will be a valuable contribution as there is currently no NrfB encoding part in the Registry and both NrfA and B are required for a functional Nrf heterotetramer. Depending on the structural and/or functional impact of the mutation, this part has the potential to be optimised by a future team.
