C2V_ArsenicCircuit
C2V_ArsenicCircuit is Cadets2Vets arsenic detection circuit. It is a 2-part circuit that combines 2 series of promoter-gene sequences on one plasmid.
The first part of the circuit (BBa_K2248001; PcArsR) contains an E. coli-derived constitutive promoter (Pc), Ribosome Binding Site, Arsenic Regulator (ArsR), and Double Terminator. This sequence allows unregulated expression of ArsR, which is a protein that will change conformation upon the binding of arsenic ions.
The second part of the circuit (BBa_K2248002; PArsRGFP) contains an E. coli-derived Arsenic Regulator promoter (PArsR), Ribosome Binding Site, Green Fluorescent Protein reporter (GFP), and Double Terminator. PArsR sequence has a binding site for ArsR. ArsR is a negative repressor and will release its transcriptional regulation in the presence of arsenic ions. The GFP reporter will turn on and fluoresce in the presence of arsenic.
The two parts were joined together using a SalI Restriction Enzyme Site.
