tyrosyl-tRNA
For incorporation of non-canonical amino acids orthogonal tRNA and aminoacyl-tRNA synthetases are needed. This pair needs to be adapted to recognize and incorporate the the non-canonical amino acid with high specifity. This part contains the tyrosyl-tRNA of Methanococcus jannaschii which is adapted to recognize the amber codon.
Usage and Biology
tRNAs are esterified with amino acids by their corresponding aminoacyl-tRNA synthetases. This part is essential for the selection plasmids to select functioning aminoacyl-tRNA snthetases from the generated library. This tyrosyl-tRNA was adapted to suppress the amber codon allowing the incorporation of non-canonical amino acids at the this particular stop codon.
