cysPUWA Sulfate Transport System cds
The cysPUWA genes encode the sulfate ABC membrane transporter in Escherichia coli. This system also transports related species such as chromate. BioBrick BBa_K2194003 contains the cds for cysP, cysU, cysW, and cysA in the context in which they appear in E. coli MG1655.
Biology and Usage
The binding protein cysP increases the affinity for sulfate uptake; genes cysU and cysW encode the integral membrane proteins. The final cds in this BioBrick is for cysA which is an ATP-binding subunit.
Figure 1 shows the effect of cysPUWA overexpression on the cells' susceptibility to Cr(VI). The lower growth rate for cysPUWA transformants in the presence of Cr(VI) indicates the higher rate of chromium import and resulting toxicity.
