TT-pLux-RBS-LuxI 2 sites-RBS-RFP-RBS-GFP-pBAD-RBS-LuxR

pLacI drives LuxR gene, producing LuxR protein for activation of pLux promoter. pLux drives LuxI gene, producing enzyme to make 3OC6, required for LuxR activation. The circuit has amplification by the LuxI enzyme and positive feedback. pLux also drives RFP gene, followed by GFP gene. The LuxI gene is modified with restriction sites for AscI and AsiSI, for introduction of frameshift mutations.
