LPE - Lactate polymerizing enzyme

This gene is a synthetic gene that is derivated from the PhaA synthase from Pseudomonas sp. 61-3. As a wild type this enzyme catalyzes the last step in the synthesis of the bioplastic polyhydroxybutyrate (PHB). However, this gene has been altered in three of its amino acids. These changes in its amino acid sequence are as follows

S325T

F395S

Q481K

These three changes all alters the active site of the enzyme, meaning that it will get an affinity towards lactyl-CoA as well. In its wild type version, this enzyme will only use (R)-3-hydroxybutyrate as a substrate, but having the enzyme with these three mutations, its substrate specificity will be moved to also include lactyl-CoA. This will change the end product that now will be P(LA-co-3HB) that is an approximately 50% PLA and 50% PHB.
