XOR gate like repressor of CI repressor expression
This part uses deoxyribozyme ribonucleases to degrade CI repressor mRNA, thereby silencing the gene. The deoxyribozyme ribonucleases are present in two copies, where expression of one is controlled by the lacI promoter and the other by the tetR promoter.
The regulatory system works like an XOR gate: Inducing the regulator with one, and only one, inducer results in silencing of CI mRNA expression. Inducing both lacI and tetR promoters results in the deoxyribozyme ribonucleases binding to each other through designed complimentary regions, stopping their mRNA degrading activities. Lack of induction of either promoter should prevent either deoxyribozyme ribonuclease from being synthesized by the cell, in turn preventing their degradation of the CI repressor.
